Two examples of nutritional pathology related to vitamins E and C deficiencies ArchiMer
Baudin Laurencin, F; Messager, J; Stephan, Gwendal.
Symptoms recalling a nutritional pathology and more precisely a process of lipid peroxydation were observed during the last years in seabass Dicentrarchus labrax and other fishes cultured in tropical marine condition, Ocyurus chrysurus and Lutjanus analis . They included dark coloration, skin ulceration, lethargy, anorexy, emaciation. Histological examinations showed hepatic fatty degenerative lesions, pancreatitis, muscular degeneration and retinal atrophy. Additional vitamins E and C in the food suppressed that pathological symptoms. The previously evoked clinical or histological signs were not achieved in spite of an increase of the level of hepatic, muscular or blood malondialdehyde and of the conjugated dienes of the perivisceral fat. In the same...

Improvement of feed intake through supplementation with an attractant mix in European seabass fed plant-protein rich diets ArchiMer
Dias, Jorge; Gomes, Ef; Kaushi, Sj.
The incorporation of an attractant mixture by substitution of an equivalent amount of basal dietary mixture was studied in terms of feed intake and growth performance in European seabass juveniles (mean initial weight : 17 g) fed plant-protein rich diets during 21 days. Three diets had as main protein sources; fish meal, or one of two soy protein concentrates (S and E). To the basal mixture of these last two diets, as well as to a corn-gluten based diet, an attractant amino acid mixture was added at a 2.5% level. Daily feed intake was measured throughout the experimental period. Feed intake and weight gain were highest in seabass fed the fish meal based diet. In those fed the soy protein concentrate diets, the addition of the attractant mixture improved...

Impact of aquaculture on the genetic structure of Mediterranean populations of Dicentrarchus ArchiMer
Bahri Sfar, Lilia; Lemaire, Christophe; Chatain, Beatrice; Divanach, Pascal; Ben Hassine, Oum Kalthoum; Bonhomme, Francois.
A phylogeographic analysis based on 6 polymorphic microsatellites was performed on 15 sea bass samples. five from Western Mediterranean, seven from Eastern Mediterranean and three coming from French aquaculture stocks. Among the eastern samples, three did not cluster according to their geographic origin but rather with the occidental group. Furthermore. a somewhat lower allelic diversity was observed within these particular samples, indicating that they originated from a limited number of progenitors of foreign origin. Among the aquaculture stocks, only one revealed a significant reduction of genetic variability, indicating that these stocks are largely outbred and open to wild fishes. The fact that the use of occidental fingerlings to seed broodstocks in...

Response to domestication and selection for growth in the European sea bass (Dicentrarchus labrax) in separate and mixed tanks ArchiMer
Vandeputte, Marc; Dupont Nivet, M; Haffray, P; Chavanne, H; Cenadelli, S; Parati, K; Vidal, Marie-odile; Vergnet, Alain; Chatain, Beatrice.
Selective breeding of European sea bass (Dicentrarchus labrax) receives a growing interest, as the estimated heritability of growth is medium to high. In this study, we compared the offspring of four groups of sea bass sires, mated with the same wild dams: wild (W), first generation of domestication (D), first generation of mass selection for length (M), first generation of PROSPER-like selection for length (P). The comparison was done both in replicated tanks (separate rearing) and in mixed tanks (mixed rearing) where sire origins were recovered by genotyping of eight microsatellite markers. Weight, length and growth rate were measured from day 238 post-fertilization (69 g mean weight) to day 611 post-fertilization (390 g mean weight). Both in mixed and...

Combined effects of dietary HUFA level and temperature on sea bass (Dicentrarchus labrax) larvae development ArchiMer
Vagner, Marie; Robin, Jean; Zambonino, Jose-luis; Person, Jeannine.
The purpose of this study was to investigate the combined effect of the incorporation of vegetable products in diet and temperature on enzymatic pathways for high unsaturated fatty acids (HUFA) desaturation in sea bass larvae. Four replicated groups were fed a low (LH; 0.8% EPA+DHA) or a high (HH; 2.2% EPA+DHA) n-3 HUFA microparticulated diet from mouth opening, six days post-hatching and were reared at 16 or 22 degrees C. The four experimental conditions (LH16, HH16, LH22 and HH22) were tested for 45 days. At the end of the experiment, body weight, total length and biomass were affected by temperature (P<0.001), while biomass as well as fresh body weight was also influenced by diet (P<0.05 and P<0.001 respectively). This always lead to the same...

Human chorionic gonadotropin induces spermatogenesis and spermiation in 1-year-old European sea bass (Dicentrarchus labrax): Assessment of sperm quality ArchiMer
Schiavone, R; Zilli, L; Vilella, S; Fauvel, Christian.
The aims of the present study were (a) to compare sperm quality (percentage of motile spermatozoa, motility duration, density and fertility after cryopreservation) between precocious and normally maturing male European sea bass Dicentrarchus labrax, (b) to examine the potential of human chorionic gonadotropin (hCG) to increase spermiation in precocious males and (c) to examine the potential of hCG to induce spermatogenesis and spermiation in non-precocious 1-year-old males. One hundred precocious and 100 non-precocious fish were each randomly divided in two groups each: control (precocious saline-treated and non precocious saline-treated) and treated (precocious hCG-treated and non precocious hCG-treated). Treated groups were administered weekly with 1000...

Dietary probiotic live yeast modulates antioxidant enzyme activities and gene expression of sea bass (Dicentrarchus labrax) larvae ArchiMer
Tovar-ramirez, D.; Mazurais, David; Gatesoupe, J. F.; Quazuguel, Patrick; Cahu, Chantal; Zambonino-infante, J. L..
The main goal of this work was to determine the effect of dietary live yeast Debaryomyces hansenii on the enzymatic antioxidative status of sea bass Dicentrarchus labrax larvae. Growth, activity and expression of the main antioxidative enzymes: catalase (CAT), glutathione peroxidase (GPX) and superoxide dismutase (SOD), and heat shock protein (HSP70) were measured in sea bass larvae at 23 and 48 days after hatching. Larvae were fed on two microdiets: group one, fed microdiet containing live yeast and the control group fed microdiet without yeast. Heat shock protein 70 showed the same expression levels in both fish larvae fed yeast and the control diet. The group fed D. hansenii showed highest growth and lower activity and expression levels of GPX and SOD...

Production de juvéniles de bar (Dicentrarchus labrax) en marais. Etude de trois séquences alimentaires de sevrage ArchiMer
Buchet, Vincent; Palvadeau, Hubert; Boussard, O..
Trials on the seabass weaning in atlantic marshes (Roux, 1993), have shown that the use of frozen prey improved this step and reduced his resulting stress. During the present study, which must confirmed these preliminary results, three alimentary sequences were tested. 1) natural zooplankton, weaning pellets, as control, 2) natural zooplankton, inert artemia (intensively produced), weaning pellets, 3) natural zooplankton, natural frozen zooplankton (harvested), weaning pellets. The achieved results show that weaning with intermediary phases (sequences 2 or 3) yield best results. Furthermore the phase with artemia yields, systematically, better results than with harvested zooplankton ; then artemia seems a excellent substitute food representing a...

Approche quantitative de la fonction de nourricerie jouée par les systèmes vasières-estuaires ArchiMer
Parlier, Emmanuel.
Coastal fish populations settle more or less frequently, at various stages, estuarine systems. Tidal marshes are one of the components of these mosaics of habitats. The first part of this work describes fish assemblages through the utilisation of these systems by fishes analysed in order to propose a revised functional classification of ecological guilds. T h i s new classification is used to compare fish communities of four estuarine systems in the western coast of France: the Mont Saint- Michel Bay, the Saint-Brieuc Bay, the Loire estuary and the Aiguillon Bay. The second part aims at characterizing the nursery function as defined by Beck et al. (2001). Acombinative approach based on gut content analysis, and stable isotope analysis shows that Yo Y s e a...

A shortcut method for sexing juvenile European sea bass, Dicentrarchus labrax L. ArchiMer
Menu, Bruno; Peruzzi, Stefano; Vergnet, Alain; Vidal, Marie-odile; Chatain, Beatrice.
The objective of this study was to develop a rapid and effective method of sexing juvenile sea bass with minimum labour and material. To this end, the gonad squash mount technique was applied along with macroscopic techniques for sexing a large number of experimental fish at the age of 215-275 days post fertilization (p.f.). At this age, 90% of the 3894 fish could be unambiguously sexed by macroscopic examination of their gonads, whereas the remaining proportion was identified using squash mount preparations (10%). The accuracy of the observations was measured up to classical histology procedures. Undifferentiated fish accounted for 0.1% of the total population only. The application of the squash mount technique holds out the prospect of considerable...

Effects of dietary tannin on growth, feed utilization and digestibility, and carcass composition in juvenile European seabass ( Dicentrarchus labrax L.) ArchiMer
Omnes, Marie-helene; Le Goasduff, Julien; Le Delliou, Herve; Le Bayon, Nicolas; Quazuguel, Patrick; Robin, Jean.
Plant-based products in fish diets are valuable protein alternatives to fishmeal for the aquafeed industry. Many plant feed ingredients contain polyphenolic compounds, including tannins, which can have beneficial or adverse effects. The tolerable threshold of ingested tannins is unknown for marine carnivorous fishes. We studied the effects of tannic acid (TA) supplementation to the diet of juvenile European seabass (Dicentrarchus labrax) by measuring growth, feed utilization and digestibility, and carcass composition. We randomly allocated groups of fish (initial mean body weight of 10.2 ± 0.7 g; n = 40 fish per tank) to 12 replicate cylindrical-conical tanks (three per treatment). The fish were assigned to one of four dietary treatments for five weeks:...

Estimation et amélioration des performances zootechniques de l'élevage larvaire de Dicentrarchus labrax et de Sparus auratus ArchiMer
Chatain, Beatrice.
By gathering specific measurement criteria and an objective analysis of field methods, the main purpose of this work is to help estimate larval Sea Bass (Dicentrarchus labrax) and Sea Bream {Sparus auratus) stock performance raised under intensive conditions. As such, it is a methodology guide zootechnicians can use, in their daily quantitative and qualitative evaluation stock issues. The first part of this work covers defining the precision of current counting methods and establishing their limitations. The quantification methods analysed, as well as the new proposed methods, relate to the major phases of larval farming, from laying to postlarval stage. Three field methods commonly used in aquaculture to quantify egg population are therefore...

Ontogenic effects of early feeding of sea bass (Dicentrarchus labrax) larvae with a range of dietary n-3 highly unsaturated fatty acid levels on the functioning of polyunsaturated fatty acid desaturation pathways ArchiMer
Vagner, Marie; Robin, Jean H.; Zambonino, Jose-luis; Tocher, Douglas R.; Person, Jeannine.
Four replicated groups of sea bass (Dicentrarchus labrax) larvae were fed diets containing an extra-high level of highly unsaturated fatty acids (HUFA) (XH; 3.7% EPA + DHA), a high level of HUFA (HH; 1.7%), a low level of HUFA (LH: 0.7%) or an extra-low level of HUFA (XLH; 0.5%) from day 6 to day 45 (experiment 1; XH1, HH1, LH1, XLH1). After a subsequent I-month period feeding a commercial diet (2.7% EPA + DRA), the capacity of the four initial groups to adapt to an n-3 HUFA-restricted diet (0-3% EPA + DHA; R-groups: XH2(R), HH2(R), LH2R, XLH2(R)) was tested for 35 d. Larval dietary treatments had no effect on larval and juvenile survival rates. The wet weight of day 45 larvae was higher in XH1 and HH1 (P<0.001), but the R-juvenile mass gains were...

Background Fully isogenic lines in fish can be developed using “mitotic” gynogenesis (suppression of first zygotic mitosis following inactivation of the sperm genome). However, genome-wide verification of the steps in this process has seldom been applied. We used ddRADseq to generate SNP markers in a meiotic gynogenetic family of European seabass (Dicentrarchus labrax): (i) to verify the lack of paternal contribution in a meiotic gynogenetic family; (ii) to generate a gene-centromere map from this family; (iii) to identify telomeric markers that could distinguish mitotic gynogenetics from meiotic gynogenetics, which sometimes arise spontaneously in mitotic gynogenetic families. Results From a single meiotic gynogenetic family consisting of 79 progeny, 42...

Individual fish rhythm directs group feeding: a case study with sea bass juveniles (Dicentrarchus labrax) under self-demand feeding conditions ArchiMer
Millot, Sandie; Begout, Marie-laure.
The long term influence of individual biological rhythms on group feed demand behaviour was investigated in European sea bass (Dicentrarchus labrax) held under controlled environmental conditions with an on-demand feeding system. The experiment was realized over 219 days with 190 fish distributed on 4 tanks. Sea bass had a mean body mass comprised between 139 g to 183 g. The number of feed demand acts by each individual was calculated daily, and the population could thus be partitioned into three categories (high-, low-and zero-triggering fish). The duration of the period that an individual held high-triggering status could vary, but was 63 +/- 16 days on average. The transition period between two highest-triggering fish in one tank was on average 4 +/- 4...

The relationship between serum protein, immunoglobulin concentrations and protein molecular weight profile (PMWP) alterations of sea bass (131.3 +/- 4.3 g) reared in sea water with sublethal concentrations of ammonia was studied over two periods totalling 116 days. During the exposure period (62 days) the first group (group 1) lived in sea water with 0.204 mg.L-1 unionized ammonia nitrogen equivalent to 12 % of the lethal concentration for 50 % of a population exposed for 96 h to ammonia (96-h LC50), whilst the second group (group 2) lived in sea water with 0.340 mg.L-1 UIA-N equivalent to 20 % of the 96-h LC50 of ammonia. Then, the two groups were left for a recovery period (54 days) in the same water as the control group (group 0). The determination of...

Is it possible to influence European sea bass (Dicentrarchus labrax) juvenile metabolism by a nutritional conditioning during larval stage? ArchiMer
Vagner, Marie; Zambonino, Jose-luis; Robin, Jean; Person, Jeannine.
The purpose of this study was to check if it is possible to influence sea bass juvenile metabolism by a conditioning of larvae from day 6 post hatching to day 45 to a low or a high HUFA compound diet (LH, 0.8% EPA + DHA and HH, 2.2% EPA + DHA) when reared at 16 or 22 °C. Following a 3-month intermediate period (at 19 °C using a commercial diet), the adaptability of the 4 initial larval groups to a HUFA experimental deprived diet (0.5% EPA + DHA) were tested at 19 °C in a 60 day-experiment (d-151–211). The four experimental duplicated conditions were ex-LH16 and ex-HH16 for the 2 groups previously reared at 16 °C (initial weight, 7.3 ± 0.5 g) and ex-LH22 and ex-HH22 for the 2 groups previously reared at 22 °C (initial weight, 11.1 ± 0.5 g). What is the heritable component of spinal deformities in the European sea bass (Dicentrarchus labrax)? ArchiMer
Bardon, Agnes; Vandeputte, Marc; Dupont-nivet, Mathilde; Chavanne, Herve; Haffray, Pierrick; Vergnet, Alain; Chatain, Beatrice.
Malformations in farmed fish are a prime research topic as it has a major impact on fish welfare and on the economical value of the product. In this study, the genetic component of spine deformities was estimated in European sea bass, a major species of Mediterranean aquaculture, with the use of a posteriori reconstruction of pedigree with microsatellites. Our population exhibited a very high malformation rate (81%),probably due to intense swimming of fish during pre-growing The heritability of global spine deformities was evaluated to be 0.21 +/- 0.04 on the underlying scale (033 +/- 0.06 for lordosis, 0.13 +/- 0.04 for scoliosis). Deformities scored by an internal or external examination turned out to be genetically the same trait (genetic...

Significance of metallothioneins in differential cadmium accumulation kinetics between two marine fish species ArchiMer

Impacted marine environments lead to metal accumulation in edible marine fish, ultimately impairing human health. Nevertheless, metal accumulation is highly variable among marine fish species. In addition to ecological features, differences in bioaccumulation can be attributed to species-related physiological processes, which were investigated in two marine fish present in the Canary Current Large Marine Ecosystem (CCLME), where natural and anthropogenic metal exposure occurs. The European sea bass Dicentrarchus labrax and Senegalese sole Solea senegalensis were exposed for two months to two environmentally realistic dietary cadmium (Cd) doses before a depuration period. Assessment of sea bass (Dicentrarchus labrax, L.) stock delimitation in the Bay of Biscay and the English Channel based on mark-recapture and genetic data ArchiMer
Fritsch, Manon; Morizur, Yvon; Lambert, E; Bonhomme, Francois; Guinand, B.
Sea bass (Dicentrarchus labrax) fisheries are becoming increasingly abundant along eastern Atlantic coastlines, especially in the ICES sub-areas IV, VII and VIII, but they are still not being managed by the European Union TAC (Total Allowed Catches) system. Scientific information about these sea bass stocks is limited, but the fishery is currently managed or modelled based upon these data, separated into the ICES divisions. Clearly, failure to manage satisfactorily the fishery may lead to a decline in stocks. We therefore investigated the structure of Atlantic sea bass populations at eight microsatellite loci in the Bay of Biscay and the English Channel and, additionally, at five selected loci in Ireland and Scotland. Genetic data showed no significant...
